Structural analysis of viral ExoN domains reveals polyphyletic hijacking events

چکیده
Nidoviruses and arenaviruses are the only known RNA viruses encoding a 3’-5’ exonuclease domain (ExoN). The proofreading activity of ExoN has played key role in growth nidoviral genomes, while this partakes suppression host innate immune signaling. Sequence structural homology analyses suggest that these proteins have been hijacked from cellular hosts many times. Analysis available sequences reveals high conservation level comparable to viral RNA-dependent polymerases (RdRp), which most conserved proteins. Two highly preserved zinc fingers present all exonucleases, arenaviral protein one finger can be identified. This is sharp contrast with reported lack ExoNs, opens possibility therapeutic strategies struggle against COVID-19.

Structural basis underlying viral hijacking of a histone chaperone complex
The histone H3.3 chaperone DAXX is implicated in formation of heterochromatin and transcription silencing, especially for newly infecting DNA virus genomes entering the nucleus. Epstein-Barr virus (EBV) can efficiently establish stable latent infection as a chromatinized episome in the nucleus of infected cells. The EBV tegument BNRF1 is a DAXX-interacting protein required for the establishment...
متن کاملComputational analysis reveals a correlation of exon-skipping events with splicing, transcription and epigenetic factors
Alternative splicing (AS), in higher eukaryotes, is one of the mechanisms of post-transcriptional regulation that generate multiple transcripts from the same gene. One particular mode of AS is the skipping event where an exon may be alternatively excluded or constitutively included in the resulting mature mRNA. Both transcript isoforms from this skipping event site, i.e. in which the exon is ei...
متن کاملViral hijacking of mitochondrial lysyl-tRNA synthetase.
The primer for reverse transcription of the human immunodeficiency virus type 1 (HIV-1) genome is tRNA3(Lys). During assembly of HIV-1 particles, tRNA3(Lys) is taken up from the host cell along with lysyl-tRNA synthetase (LysRS), the tRNA binding protein that specifically aminoacylates the different tRNA(Lys) isoacceptors. In humans, the cytoplasmic and mitochondrial species of LysRS are encode...

متن کاملStructural and functional analysis of Nup133 domains reveals modular building blocks of the nuclear pore complex
Nucleocytoplasmic transport occurs through nuclear pore complexes (NPCs) whose complex architecture is generated from a set of only approximately 30 proteins, termed nucleoporins. Here, we explore the domain structure of Nup133, a nucleoporin in a conserved NPC subcomplex that is crucial for NPC biogenesis and is believed to form part of the NPC scaffold. We show that human Nup133 contains two ...
